I think Regina thought that because Ava and Nicholas were trying to leave, a "bad thing" would happen. Hopefully that bad thing would kill Emma. If not, Emma could at least suffer and feel guilty for the harm it did the kids.
However, I'm not completely sold on that, I admit. It seems to slip-shod, like leaving too much up to chance (after all, Ashley wasn't severely injured). Then again, that did seem to be her tactic with Kathryn, so maybe that was the plan.
